I've built up an HTPC with MP on it, but I want to use it for more than just my TV. I have an old hand-me-down rear-projection big screen with only CATV, Composite and S-video ins. My output to the TV options are S-Video. My 22" wide screen monitor is dvi or vga. What I'm wanting to do is have the case next to the tv, but have my monitor, keyboard and mouse on a desk in another room (likely separated by a ceiling/floor), and be able to use MP on the TV while playing a game on the monitor. Can it be done?

It's definitely possible. The only real limitation you're going to run into will be your video card (whatever it may be). If you're wanting to run an intensive game on one screen and have MP run smoothly on your secondary monitor, you'll need a good video card. Also, you'll want to stay under (well under) 100ft when running an s-video/composite cable. I'm not sure how far away you're talking about, but that might be another problem you'll face. Other than those two potential problems, you shouldn't have too much trouble doing what you want to do.
