ok, i delete this famous file, it runs. but only for few hours... should i delete this file per Autostart ? i think that can be not the right thing, or?

No you should not delete the file every time you start MediaPortal / Your PC.
You should just delete it once, as to clear the folder database (creating a new one).
After this you may experience folders to open slow the first time (anyone please correct me if I'm wrong) but not after that.
What do you mean by "it runs. but only a few hours" ? In case Mediaportal crashes you should have a crash report (logs) available that you can upload. I do suggest you start a thread regarding that issue as this is about MP being slow not crashing.

I think he means that mediaportal runs smooth for a few hours, and becomes slow again after that.
