Same problem over here. On Media Portal 1.0 the system detected the TV Server Configuration as requiring Administrator priviledges. With 1.0.1 it can run in User Mode, but in this mode it will not work. I don't know how to fix this, except forcing it to run as an Administrator with the context menu.
Update: if you open the properties of the shortcut there's a button with advanced options that allows you to run it as an administrator every time. Enable this and that'll fix the problem. There's no need to disable UAB or tweak permissions (I recommend to leave the default settings).
